IN-VITRO MASS PROPAGATION OF SUGARCANE FROM SPINDLE LEAVES

Plant tissue culture offers the best methodology through micro propagation of plants for quality and phytosanitary planting material at a faster rate in a shorter period. This study was conducted to find a composition of the combination of Plant Growth Regulator (PGR) to effective regeneration method for mass propagation of sugarcane using sheath leaves as explants. The explants of sugarcane was cultured on MS medium supplemented with combination of auxin and cytokinin at various concentration, as follows: NAA 1 mg.L-1 + kinetin 1 mg.L-1 , NAA 1.5 mg.L-1 + kinetin 1 mg.L-1, NAA 2 mg.L-1 + kinetin 1 mg.L-1 , NAA 1 mg.L-1 + kinetin 1.5 mg.L-1 , NAA 1 mg.L-1 + kinetin 2 mg.L-1 , NAA 1.5 mg.L-1 + kinetin 1.5 mg.L-1 , NAA 2 mg.L-1 + kinetin 1.5 mg.L-1 , NAA 1.5 mg.L-1 + kinetin 2 mg.L-1 dan NAA 2 mg.L-1 + kinetin 2 mg.L-1 . The research was design in a random completed design which comprised of four replicates and every replicate consisted of two observation material units. Results revealed that combination of NAA 2 mg.L-1 + kinetin 1 mg.L-1 and NAA 1.5 mg.L-1 + kinetin 2 mg.L-1 were the best combination for roots and leaves multiplication respectively. Meanwhile, combination of NAA 2 mg.L-1 + kinetin 1.5 mg.L-1 and NAA 1 mg.L-1 + kinetin 2 mg.L-1 were the best combination of concentration to achieve the fastest growing of roots and shoots respectively.
